I am wondering if anyone knows of any issues that I might encounter replacing the sear housing block assembly on a 2016 M&P9L Pro C.O.R.E with this one available from Midwest Gun Works:

Smith & Wesson M&P 9mm / .40 S&W Sear Housing Block Assembly, Compact & Standard

The purpose would be to add a manual thumb safety.

I know that I would need to cut notches in the frame to accommodate the safety, but I am wondering if there are any other issues that I was not aware of that might cause a problem. Thanks in advance for assistance.

Can't do it unless you have the right frame. There's more to it than the notches. All the M&P45 frames are designed for a thumb safety, but the black 9mm/40S&W/357SIG have a different frame for the safety version. I haven't seen a tan M&P frame that wasn't safety capable.
